Shes been my favorite of the 6 since the beginning.
Fluttershy is cutest.
Pinkie has the most entertaining behavior.
Rainbow Dash is the coolest.
Twilight is adorkable, yet highly intellectual and respectable. Some would even say relatable.
Applejack is the most all around strong and capable.
Rarity is the prettiest and most graceful.
Of course, I've got an enormous and soft spot for cute things, so my favorite is Fluttershy.
PS: Now in case somepony gets upset because I called Twilight a dork, allow me to explain my interpretation of the term; Adorkable describes someone who is socially awkward/ignorant/oblivious to the point that it's charming and cute. Weird to be around if you're a stranger, but you'd love her if you get to know her.
